Ondansetron is indicated for the prevention and treatment of nausea and vomiting associated with moderately and highly emetogenic chemotherapy, radiotherapy, and post-operative nausea and vomiting (PONV). In paediatric practice, it is widely used for acute gastroenteritis-related vomiting to allow oral rehydration. It is indicated from 6 months of age in many guidelines for gastroenteritis-related vomiting under medical supervision.
When the gut or certain areas of the brain sense irritation — from an infection, chemotherapy, or surgery — they release serotonin, which activates the vomiting reflex. Ondansetron blocks the specific serotonin receptors (5-HT3 receptors) that trigger nausea and vomiting, interrupting this signal and quickly settling the urge to vomit, allowing the child to tolerate fluids and food.
Ondansetron is a selective antagonist of serotonin (5-HT3) receptors located on vagal afferent nerve terminals in the gastrointestinal tract and in the chemoreceptor trigger zone (CTZ) of the area postrema in the brainstem. Emetogenic stimuli (infection, cytotoxic drugs, surgical trauma) trigger enterochromaffin cells to release serotonin, which activates 5-HT3 receptors and initiates the vomiting reflex via the vomiting centre. Ondansetron competitively blocks these receptors, preventing serotonin-mediated neural activation and inhibiting both peripheral (gut) and central (CTZ) initiation of nausea and vomiting.
Ondansetron was originally developed in the 1980s to manage the severe nausea caused by cancer chemotherapy. Its success was so remarkable that it revolutionised the supportive care of cancer patients, and it has since become one of the most widely used antiemetics in the world across many different clinical settings.
Ziaset Drop should be given as prescribed by your doctor. It can be given with or without food. The drop form allows it to be measured and administered directly or mixed with a small amount of liquid. Use only as directed — do not repeat doses more frequently than prescribed.

Ondansetron is generally well tolerated. Common side effects may include headache, constipation, or a feeling of warmth or flushing. Dizziness may occasionally occur. These are usually mild and resolve quickly. Tell your doctor if any side effect is severe or persistent.
If vomiting stops, the infection causing it is cured.
Ondansetron relieves the symptom of vomiting but does not treat the underlying infection or illness. Treatment of the cause and maintaining hydration with ORS remains essential.
Antiemetic medicines are not needed — children will stop vomiting on their own.
While mild vomiting may self-resolve, persistent vomiting in children can quickly lead to dangerous dehydration. Ondansetron helps control vomiting so children can accept oral rehydration fluids, which is critically important for recovery.
