Mechanism of Action

Pantoprazole is a substituted benzimidazole that irreversibly inhibits the hydrogen-potassium ATPase (H+/K+ ATPase) enzyme — the gastric proton pump — on the secretory surface of gastric parietal cells. After parenteral administration, pantoprazole is absorbed rapidly and concentrated in the acidic environment of the parietal cell canaliculi, where it is converted to the active sulphenamide form that covalently binds to cysteine residues (particularly Cys-813 and Cys-822) of the H+/K+ ATPase, producing irreversible acid suppression that lasts until new pump proteins are synthesised (24–48 hours).
