Ziadrene Tablet combines Drotaverine HCl 80 mg and Mefenamic Acid 250 mg to provide targeted relief from spasmodic pain — the cramping, colicky pain caused by smooth muscle contractions in the abdomen, uterus, or urinary tract. Drotaverine relaxes smooth muscle spasm while Mefenamic Acid reduces the inflammatory prostaglandins that trigger spasm and pain.
This combination is particularly favoured for the management of dysmenorrhoea (menstrual cramps), abdominal colic, and biliary or renal colic. By addressing both the spasm and the prostaglandin-driven inflammation simultaneously, Ziadrene provides faster and more effective relief than either component alone.

Ziadrene is indicated for the symptomatic treatment of smooth muscle spasm-associated pain in adults, including primary dysmenorrhoea, gastrointestinal colic (spastic colon, biliary colic), ureteric or renal colic, and postoperative abdominal cramping. Mefenamic Acid is particularly well established in the management of primary dysmenorrhoea due to its ability to inhibit uterine prostaglandin synthesis.
Drotaverine in Ziadrene relaxes tight, cramping smooth muscles — the type of muscle found in the uterus, gut, and urinary tract — by reducing the chemical signals that keep them contracted. Mefenamic Acid works by blocking the production of prostaglandins, the hormone-like substances that cause both the cramping and the pain of conditions like period pain. Together, they stop both the source of cramping and its painful effects.
Drotaverine is a phosphodiesterase-4 (PDE-4) inhibitor that selectively blocks the enzyme responsible for degrading cyclic AMP (cAMP) in smooth muscle cells. Elevated cAMP activates protein kinase A, which phosphorylates myosin light chain kinase (MLCK), preventing myosin-actin interaction and causing smooth muscle relaxation — similar in effect to papaverine but with greater smooth muscle selectivity and no cardiac effects. Mefenamic Acid is a non-selective COX-1 and COX-2 inhibitor and also a direct antagonist of prostaglandin receptors (EP1 and EP3), providing both synthesis inhibition and receptor-level blockade of prostaglandin action — a dual mechanism particularly effective for prostaglandin-mediated uterine pain in dysmenorrhoea.
Mefenamic Acid, a key component of Ziadrene, is one of the few NSAIDs that not only blocks prostaglandin production but also directly blocks prostaglandin receptors. This dual mechanism gives it a particularly strong effect against prostaglandin-driven pain like primary dysmenorrhoea, which is why it has remained a preferred choice for period pain for decades.
Ziadrene is taken as directed by your doctor, typically 2–3 times daily with food to reduce stomach irritation. For period pain, it is often most effective when started at the first sign of cramping or the day before menstruation is expected. Always follow your doctor's prescribed regimen.
