Ondansetron 4 mg is indicated for the prevention and treatment of nausea and vomiting associated with moderately and highly emetogenic chemotherapy, radiotherapy, and post-operative settings. It is also used off-label for nausea associated with acute gastroenteritis and hyperemesis gravidarum under medical supervision.
Ondansetron blocks serotonin (5-HT3) receptors in the gut and in the brain's vomiting centre. When these receptors are blocked, the signals that trigger nausea and vomiting are interrupted — providing fast and effective relief from these symptoms.
Ondansetron is a selective 5-HT3 receptor antagonist. During emetogenic stimuli (chemotherapy, surgery, gastrointestinal irritation), enterochromaffin cells in the gut mucosa release serotonin (5-HT), which activates vagal afferent nerve fibres via 5-HT3 receptors and stimulates the chemoreceptor trigger zone (CTZ) in the area postrema. Ondansetron competitively blocks 5-HT3 receptors at both central and peripheral sites, preventing the initiation and propagation of the nausea and vomiting reflex.
